Objective:
The locus coeruleus (LC) innervates the cerebrovasculature and plays a crucial role in optimal regulation of cerebral blood flow. However, no human studies to date have examined links between these systems with widely available neuroimaging methods. We quantified associations between LC structural integrity and regional cortical perfusion and probed whether varying levels of plasma Alzheimer’s disease (AD) biomarkers (Aß42/40 ratio and ptau181) moderated these relationships.
Participants and Methods:64 dementia-free community-dwelling older adults (ages 55-87) recruited across two studies underwent structural and functional neuroimaging on the same MRI scanner. 3D-pCASL MRI measured regional cerebral blood flow in limbic and frontal cortical regions, while T1-FSE MRI quantified rostral LC-MRI contrast, a well-established proxy measure of LC structural integrity. A subset of participants underwent fasting blood draw to measure plasma AD biomarker concentrations (Aß42/40 ratio and ptau181). Multiple linear regression models examined associations between perfusion and LC integrity, with rostral LC-MRI contrast as predictor, regional CBF as outcome, and age and study as covariates. Moderation analyses included additional terms for plasma AD biomarker concentration and plasma x LC interaction.
Results:Greater rostral LC-MRI contrast was linked to lower regional perfusion in limbic regions, such as the amygdala (ß = -0.25, p = 0.049) and entorhinal cortex (ß = -0.20, p = 0.042), but was linked to higher regional perfusion in frontal cortical regions, such as the lateral (ß = 0.28, p = 0.003) and medial (ß = 0.24, p = 0.05) orbitofrontal (OFC) cortices. Plasma amyloid levels moderated the relationship between rostral LC and amygdala CBF (Aß42/40 ratio x rostral LC interaction term ß = -0.31, p = 0.021), such that as plasma Aß42/40 ratio decreased (i.e., greater pathology), the strength of the negative relationship between rostral LC integrity and amygdala perfusion decreased. Plasma ptau181levels moderated the relationship between rostral LC and entorhinal CBF (ptau181 x rostral LC interaction term ß = 0.64, p = 0.001), such that as ptau181 increased (i.e., greater pathology), the strength of the negative relationship between rostral LC integrity and entorhinal perfusion decreased. For frontal cortical regions, ptau181 levels moderated the relationship between rostral LC and lateral OFC perfusion (ptau181 x rostral LC interaction term ß = -0.54, p = .004), as well as between rostral LC and medial OFC perfusion (ptau181 x rostral LC interaction term ß = -0.53, p = .005), such that as ptau181 increased (i.e., greater pathology), the strength of the positive relationship between rostral LC integrity and frontal perfusion decreased.
Conclusions:LC integrity is linked to regional cortical perfusion in non-demented older adults, and these relationships are moderated by plasma AD biomarker concentrations. Variable directionality of the associations between the LC and frontal versus limbic perfusion, as well as the differential moderating effects of plasma AD biomarkers, may signify a compensatory mechanism and a shifting pattern of hyperemia in the presence of aggregating AD pathology. Linking LC integrity and cerebrovascular regulation may represent an important understudied pathway of dementia risk and may help to bridge competing theories of dementia progression in preclinical AD studies.

The dammed river dolphins of Brazil: impacts and conservation

A major threat to river dolphins in Brazil is habitat loss and fragmentation as a result of the construction of hydroelectric dams. We examine the degree of overlap between the distributions of two river dolphin species (boto Inia geoffrensis and tucuxi Sotalia fluviatilis) and hydroelectric projects and provide an initial quantification of impacts. The locations of dams were overlaid onto a map of the known distributions of the dolphins. Two types of impacts were evaluated: population fragmentation and effects on prey. For each of these a binary score (for absence or presence) was assigned for each dam. The sum of all impact scores for each dam, in each of the three watersheds in which dolphins are found, including realized and potential impacts, was calculated as the impact index. Thirteen dams affect the distribution of I. geoffrensis (as could a further three dams under construction and seven planned), and three dams affect S. fluviatilis (as could three under construction and two planned). Overall, the impact index was higher for I. geoffrensis than for S. fluviatilis. The Araguaia–Tocantins and North Atlantic watersheds have higher impact index values than the Amazon watershed. Although some impacts of damming could not be considered in this study, our approach was a rapid initial risk assessment that provided an overall understanding of how river dolphins are and may be affected by hydroelectric projects in Brazil. Our method may be applicable to other freshwater cetaceans and species facing similar threats when information about impacts is incomplete.

Summary
Currently pregnant women in the industrialized parts of the world expect to have an ultrasound of the fetus. There is no clear evidence that ultrasound will decrease perinatal morbidity or mortality in low-risk women [1]. However, the mother's perception is that she will find out if the fetus is “normal” and what the gender is. Often she will bring a partner and/or family members.
Universal ultrasound imaging of all pregnancies has not been officially endorsed, particularly in lowrisk women; however 60–70% of women develop a medical indication resulting in a prenatal ultrasound [2]. Several bodies (the American Institute of Ultrasound in Medicine [AIUM], the American College of Obstetrics and Gynecology [ACOG], and the American College of Radiology [ACR]) agree on the indications for ultrasound in pregnancy and the minimum requirement of images, regardless of who performs the studies: radiologists or obstetricians.
Prenatal ultrasound may improve specific pregnancy outcomes indirectly. For example, ultrasound may: (1) identify congenital anomalies to allow for the option of termination or appropriate delivery or management at specialized centers; (2) accurately date the gestation in case of preterm delivery; (3) best recognize multiple gestations, as well as significant growth disturbances.

Atomic layer deposition (ALD) is ideal for applying precise, conformal coatings over nanoporous materials. We have recently used ALD to coat two nanoporous solids: anodic aluminum oxide (AAO) and silica aerogels. AAO possesses hexagonally ordered pores with diameters d∼10 nm and thicknesses L∼70 microns. The AAO membranes were coated by ALD with successive layers of A1203, TiO2 and V2O5 to fabricate catalytic membranes. SEM, TEM and EDAX analysis of the membranes demonstrate that the ALD layers uniformly coat the extremely high aspect ratio (L/d∼104) AAO pores. These catalytic membranes show remarkable selectivity in the oxidative dehydrogenation of cyclohexane. Additional AAO membranes coated with ALD Pd films show promise as hydrogen sensors. Silica aerogels have the lowest density and highest surface area of any solid material. Consequently, these materials serve as an excellent substrate to fabricate novel catalytic materials and gas sensors by ALD. In this study, both thin film and monolithic aerogels were coated by ZnO ALD and the properties of the aerogels were investigated as a function of the coating thickness.

We have demonstrated a novel cyclosulfation chemistry for the functionalization of C60 Molecules and a new route to the synthesis of fullerol. The cyclosulfation reaction of C60 was performed in neat fuming sulfuric acid at 55–60 °C under N2. Hydrolysis of these derivatives in the presence of water at 85–90 °C or in aqueous NaOH solution at ambient temperature gave the corresponding polyhydroxylated fullerenes (fullerols) in high yield. Various spectroscopie methods were used to resolve the chemical structure of fullerol. An average of 10 to 12 hydroxyl addends was found in fullerols that can be correlated to the structure of polycyclosulfated fullerene precursors containing 5 to 6 cyclosulfate units. The cyclosulfation chemistry is, presumably, initiated by an one-electron oxidation reaction on C60 Molecules, followed by the attack of nucleophilic anionie sulfate species onto the resulting cationic C60 radical intermediates. Further electron oxidation and intramolecular cyclization of the resulting hydrogen sulfated C60 yielded the desired polycyclosulfated C60 products.
Proliferation and Interactions of Several Cell Types Encapsulated Within Dense (Non-Porous) Protein-Permeable Polyurethane Membranes

Protein-permeable dense (non-porous) urethane membranes have been evaluated for in vitro cell culture, and in vivo cell encapsulation. Polyurethane membranes were designed to exhibit permeability to proteins, gases, and nutrients without the existence of pores. The membranes are non-cytotoxic, angiogenic, and permeable to gases, nutrients, secretagogues and cell products via purely concentration-driven transport. Non-anchorage and anchorage dependent cells were grown encapsulated within the membrane and with the membrane as a growth substrate. Several non-anchorage dependent cell types proliferated within the membrane both in-vitro and in-vivo. Anchorage-dependent cells were grown on the membranes as a substrate. Encapsulated cells have been maintained in culture for up to six months with nutrients supplied only by the external media. Immuno-isolation has been demonstrated with cells implanted into murine hosts. Explants of membrane encapsulated cells exhibited a high degree of vascularization, with little or no fibrous tissue. The ability to support cell growth and function, and the ability to protect xenogenic cells from immunologic rejection suggest that the membranes would be useful in the construction of hybrid artificial organs, devices for cell transplantation, and substrates for cell and tissue culture.
Testosterone, sex hormone-binding globulin, calculated free testosterone, and oestradiol in male vegans and omnivores

Total testosterone (T), total oestradiol (E2) and sex hormone-binding globulin (SHBG) concentrations were measured in plasma samples from fifty-one male vegans and fifty-seven omnivores of similar age. Free T concentration was estimated by calculation, in comparison with the omnivores, the vegans had 7% higher total T (P = 0.250), 23% higher SHBG (P = 0.001), 3% lower free T (P = 0.580), and 11% higher E2 (P = 0.194). In a subset of eighteen vegans and twenty-two omnivores for whom 4 d diet records were available, there were statistically significant correlations between T and polyunsaturated fatty acids (r 0.37), SHBG and fat (r 0.43 for total fat, 0.46 for saturated fatty acids and 0.33 for polyunsaturated fatty acids), and SHBG and alcohol (r–0.39). It is concluded that a vegan diet causes a substantial increase in SHBG but has little effect on total or free T or on E2.
